Hello again, another question - I notice in your diagrams you show the grin lens vertical. But the bone and CA1 are angled at about 15 degrees. Do you recommend keeping the implant vertical or at the 15 degree or so angle given by the bone, which I think might approximate the CA1?

Hi Brendon, for CA1 we keep the GRIN lens vertical (although we don't ensure that it is perfectly vertical). Generally, we have found that when you push the GRIN lens down, it flattens the hippocampus a little bit and the tissue will actually fill up to the lens, giving you pretty even imaging in your field of view. The damage is pretty minimal because the corpus callosum insulates it from the lens. You can certainly try to aspirate and insert the lens at an angle but we never found a need for it.

For other brain regions like cortex we usually tilt the lens to accommodate the curvature of the tissue.

For the most part there aren't any issues with using a tilted lens as mice can deal with the tilted microscope pretty well (up to ~30 degrees I would say with no issues).
