Surgery and Baseplating / Re: nothing but gray

@kjlawlor

THe people I know have tried nucleus accumbens and ventral pallidum in rats with virtually no success. However, two different people appear to get fine cells when implanting a lens in prelimbic cortex (~3-4.0mm DV), which is what I plan to target. The bleeding/inflammation definitely seems to be a big factor, their successful protocols use pre-op dexamethasone and for 3 days post-op. It is unclear if the deeper regions are nearly impossible in rats becuase of the inflammation but hopefully someone will figure it out soon.

Kristen,

What brain region are you targeting?

From what I have heard from people trying to image deep brain regions in rats (here at MUSC) using the Inscopix system is that it is very difficult and they have been unsuccessful. They have got good cells in mPFC but it seems like deep targets like NAc and VP don't yield good cells, not sure of the theory behind it, perhaps due to more inflammation in rats.

Data Analysis / Results from MIN1PIPE Analysis Pipeline?
« on: July 26, 2018, 12:36:36 AM »
Hi All,

I am curious whether anyone has tried using the new MIN1PIPE pipeline (https://www.sciencedirect.com/science/article/pii/S221112471830826X?via%3Dihub#app2) in Matlab for analyzing miniscope recordings?

I'm coming from Python and have been trying to use the CNMF-E analyses from CaImAn and MiniscoPy. Interested to see whether MIN1PIPE provides significant benefits over the prior methods.

Any insight is appreciated.

Hi All,

I am curious what methods people are using to hold the GRIN lens while driving it into the brain. I'm familiar with the heat shrink serrefine method from the Resendez protocol, but I figured someone might have a newer, more clever design.

Alternatively, has anyone had luck using the lens holder designs from other providers such as the ONE core (neuroprobeamajiger), Doric, etc.?

Thanks for your insight.

General Discussion / Metal rods for Rat Protective Cone?
« on: June 22, 2018, 09:15:37 PM »
Hey All,

I am curious if anyone has found a good iron rod or wire to hold together the two pieces of the 3D-printed protective cone for rats?

If so could you provide the specs or a link for purchase.

Thanks!
