In my opinion the most important consideration here is to minimize the excitation of ChR2 when exciting jRCeco1a. For this reason I think 49306 is the best fit.

As for picking an LED. You ideally want to pick the one that excited jRCeco1a the most when passing through your filter. This means you have to take into account both the filter band as well as the excitation spectra. I think the amber or PC-amber would work just fine.

Hi Don,
Can you upload a picture of the problematic PCB? There are 2 possible problems that could cause what your are experiencing:
  • If you are using the newest CMOS PCB version, you need to make sure to add a little solder ball across the 2 half circuits on the bottom of the PCB to short the 2 pads together. Look here, http://miniscope.org/index.php?title=System_Assembly, under the section titled "Soldering the solder-jumper on the bottom of the CMOS PCB (v3.2)".
  • The other possible cause is a damaged LED or slightly shorted LED wires or some short around the LED driver circuitry (left over flux residue can cause this). I know you tried a brand new LED and LED wires but you might want to try 1 more time.

Hi,
This definitely looks repairable but will require a bit of finesse. Under a steroscope you will want to straighten out all the bent traces on the PCB. It looks like 6 of the 10 traces are fully intact and just need to be gently bent back into place. For the other 4, you want to clean these up as much as possible and scrape off a millimeter or two of solder mask on the trace. Finally, you want to epoxy a new USB connector in place. You can solder the 6 intact traces directly to the USB connector. The other 4 will likely need you to use a very thin, single core wire to connect the USB connector pins to the traces. I would use wire wrapping wire, something from here https://www.digikey.com/products/en/cables-wires/wire-wrap/470.

This has happened to a couple of our DAQ PCBs over the past few years. If the PCB traces running up to where the USB connector is support to be mounted aren't too damaged you can probably repair this. Can you upload a close up picture of the USB connector part of the PCB?

Hi KK Ji,
This sounds like an issue with the coax cable solder joints on the CMOS PCB or the Coax2SMA PCB. I would suggest desoldering the coax cable completely and either soldering on a new one or trimming the ends of the old coax cable and re-solder it back.

Hi Tycho,
We haven't tried out GoFoton lenses in conjunction with prisms yet but I imagine they should work well. If you are interested you can contact Walter at GoFoton (walter.boyles@gofoton.com) and request custom 1mm diameter CLH lenses. You will just have to provide him with the new front working distance of the lens which will be their standard working distance plus the size of your prism. So for a 1mm x 1mm prism you would just ask them to cut some GRIN lenses to increase the front working distance by 1mm.

Please update this thread with your progress. We have found that some cheap USB3 cables perform poorly with the Miniscope system. Generally aim for higher quality cables that are no more than 5 feet in length.

Hi Honi,
Moving the focus slider up (away from the body of the scope) shifts the focal plane shallower in the brain. Using a shorter focal length Achromatic lens does the same thing. So if you find yourself needing to image less deep into the brain you can switch to a shorter focal length achromatic lens. Hope this answers your question.

Yes, we at UCLA have developed a wire-free version of the Miniscope. This system is battery powered and records to on-board memory. We are in the process of final testing and design tweaks. We plan on openly sharing this system within 6 months.
