Thank you for the information! I am targeting a deep brain region, the VPM at about 5.5 mm, is that what you were referring to as VP? I am still unsuccessful so I am somewhat comforted to know this is not so straightforward as a shallower region. Have you guys tried any methods to reduce inflammation? I plan to try giving dex pre-op for 5 days to see if this can reduce inflammation. In parallel, I will try a shallower region to at least get some good results hopefully.

I see you are also experiences differences between the mice and rats. As I am unfamiliar with your region's surgery, I cannot comment on the images you provided but I wish you the best of luck!

Hi Daniel,

Thank you so much for the reply. I have reached out to Tad Blair's lab and they have been very helpful regarding the viruses and other information. They did indeed use AAV9.

I bought the virus from addgene as well, https://www.addgene.org/51085/
So far, the histology shows that the virus expresses, however we have had to dilute it 1:10 but we are still experiencing over-expression so we may need to dilute even further.
We are using the relay lens from inscopix (8.4mm), and the objective lens from Edmund Optics, 5mm Dia. x 12.5mm FL, MgF2 Coated, Achromatic Doublet Lens.
We have not successfully seen any dynamic cells, and I believe that may be either due to the virus or possibly the lens implantation process. We are still troubleshooting to figure out how to improve. Hope your second surgery turns out well!

Hi,

I am also trying to get my imaging in Rats to work. I have implanted 10 rats using two different viruses, and have yet to see anything. I am using the two lens relay system, with the implanted lens 8.4mm long for a region that is 5.5mm deep. I am wondering if there are any changes in the protocol for rats. Also, do you know people who have successfully imaged rats using the miniscope in such deep regions?

Does the miniscope team have a list of publications that are using the miniscope system?

Hi Honi,

I am also working in rats and I have had some issues. Have you been able to get it working? If so, did you make any changes to the protocol?
Also if you don't mind me asking, are you using the objective-relay 2 lens configuration? The area I am trying to image is at a depth of 5.5mm so I am wondering if anyone has had success doing deep brain imaging in rats. Thanks!

DAQ Software/Firmware / Ms Cam Error Only When Closing Camera

Hi,

I seem to have an error with msCam but only when I attempt to close the msCam window. When the msCam window is open, it is a small, square window (seen in 'scope error 2'). When I attempt to close the msCam window, another screen pops up (also labeled msCam), opens and immediately becomes unresponsive (as seen in 'ms cam error' attachment). The program stops responding entirely and eventually I get a windows error box saying MiniScopeControl is not responding and I am forced to close the entire program. However, before trying to close the msCam window I can run the camera for as long as I want without it timing out or causing any errors. The error only occurs while closing out. Have you seen this error before or know any solutions to this issue? Thank you for the help!
