Hi,
we have two setups running experiments in two different locations. In one we use a Lenovo thinkstation that causes no problems whatsoever, however in the other we used a lenovo Laptop that had bad connection quality and after some issues with incompatible dell workstations we decided not to chance it anymore and want to get a Mac Book Pro. Also we need the mobility/space, otherwise we'd just get another Lenovo Thinkstation.

Does anyone know if the now USB-less MacBook pros (Thunderbolt 3) are still working with the Miniscope Software? I doubt that there'll be issues, but I'd rather not not find out the hard way.

DAQ Software/Firmware / Running 2 DAQ Boards on the same Workstation

Has anyone tried to run two DAQ boards (and miniscopes) on the same PC?

We thought about using a separated USB3 controller and two seperate ssds to save the data.
Buying two workstations is not an issue, however space is....

how does the field of view depend on the Achromat used?
We noticed that it changes size, depending on the focal length.

Hi,
we would like to combine jRGeco1a recordings with ChR2 excitation and are currently looking for appropriate filter sets from Chroma. Based on a previous discussion (http://miniscope.org/board/index.php?topic=393.0) on this board we picked three potential candidates with the following emission/excitations specs:

Now these are all very similar, however there are two points that we are currently still trying to sort out.
The first is that we would obviously like to avoid exciting the ChR2 along with the jRGECO1a. ChR2 excitation drops to its minimum at 550nm which means that if it is necessary to block all ChR2 excitation by the green LED, only set #49306 is an option. Does anyone here have any experience on how easily ChR2 will be unintentionally excited at the 'edges' of its spectrum?

Secondly, depending on the set of filters, different LEDs will have to be used. Luxeon currently has 4 colors that seem to have appropriate spectra.

Based on the spectra drawn in the datasheet and the three filter sets we picked, Green is probably not going to work. Lime, Amber and PC-Amber should work, however depending on the filter set some will probably require higher LED power than others to get the similar results, depending on the filter sets used.

@Daniel: Does that make any sense so far or are we overthinking here?

@anyone else: Any input to this is highly appreciated. As soon as we can provide more informartion on the technicalities we'll post them.

While it's pretty expensive, it also works the best so far.
However we do not need 1000 feet.....

The miniscope image can be displayed in Bonsai like any other webcam, however it is not possible to control any of the settings.

Hi Daniel,
we are using a software called Bonsai for some of our experiments involving Behaviour (it's a great tool for anything tracking related)
and I wondered how one could combine the two. (https://bitbucket.org/horizongir/bonsai)
The current solution is to use an ardunio, which doesn't really make sense given that the Miniscope DAQ Software and Bonsai usually run on the same PC.

Are you aware of anyone working on this?

Hi Daniel,
I see your point. We'll stick to 5 fps or higher then.
The idea is to look at astrocyte calcium dynamics, which are very slow.
Hence we figured we could cut on the amount of data produced by lowering the framerate further.

We wondered if it is possible to acquire images at frame rates below 5Hz?
